骆驼蓬内生细菌多样性与抗菌活性研究

以塔里木盆地药用植物骆驼蓬为研究对象,分离内生细菌,探究其内生细菌多样性并检测抑菌活性,发掘骆驼蓬内生菌物种资源和应用潜力。选取骆驼蓬根、茎、果实进行表面消毒,采用7种培养基进行分离培养,获得72株细菌,经16S rDNA鉴定,分布于7个目、15个科、17个属,其中优势菌属为芽孢杆菌属。以引起果树病害的解淀粉欧文氏菌为靶标,采用平板对峙法和孔碟法对骆驼蓬内生细菌进行抑菌活性筛选,获得9个活性菌株,其中芽孢杆菌属(Bacillus)4株、大洋芽孢杆菌属(Oceanobacillus)、微小杆菌属(Exiguobacterium)、链霉菌属(Streptomyces)、农杆菌属(Agrobacterium)、亮杆菌属(Leucobacter)各1株。本研究的开展为塔克拉玛干沙漠特殊环境药用植物内生菌资源的开发和农用微生物药物的开发奠定了基础。

Abstract:

In this study, the endophytic bacteria were isolated, the diversity of endophytic bacteria was investigated, and the antibacterial activity was detected, so as to explore the species resources and application potential of endophytic bacteria in Peganum harmala,a medicinal plant in the Tarim Basin. The roots, stems and fruits of P. harmala were selected for surface disinfection, and 7 kinds of media were used for isolation and culture. 72 strains of bacteria were obtained, which were identified by 16S rDNA and distributed in 7 orders, 15 families, 17 genera, among which the dominant bacteria genus was Bacillus. In this study, we screened the endophytic bacteria of P. harmala by plate and plate methods, and obtained 9 active strains, including 4 Bacillus, 1 Oceanobacillus, 1 Exiguobacterium, 1 Streptomyces, 1 Agrobacterium and 1 Leucobacter. This study laid a foundation for the development of endophytic bacteria resources of medicinal plants in special environment of Taklimakan Desert and the development of agricultural microbial drugs.
